Outline of Annual Research Achievements

加齢や各疾患でいかに概日時計が乱れるのかは未解明な部分が多く、その作用機序の解明が本研究の目的である。本国際共同研究チームは、これまでに視床下部にある概日時計中枢に着目した解析を進めてきた。特に加齢や疾患における概日時計中枢の神経活動変化を捉えるべく、UCLAにて、UCLA miniscopeをベースに、概日時計中枢のイメージング手法の確立を新規に目指すことをメインの渡航目的とした。老化促進モデルマウス、アデニン誘発性慢性腎臓病モデルマウス、カフェイン投与による概日リズム位相後退モデル、概日機能障害を伴う双極症モデルマウス、時差ボケによる性周期障害モデルなどについて、病態の解明だけでなく、ミトコンドリア機能低下、神経炎症をターゲットに、概日時計変調のメカニズム解明を目指した。2020、2021年度はコロナ禍で研究が思うように進まなかったが、オンラインミーティング等を通じて、研究手法の確立を目指した。2022年度は、UCLA miniscopeを用いた線条体シングルセルカルシウムイメージングの実験系を確立することができた。また、ライデン大学に1ヶ月間修士課程の学生を派遣し、UCLA miniscopeを用いたイメージング技術やAAVウイルスの新規注入方法について学ぶことができた。2023年度-2024年度にかけて、明治大学にて、UCLA miniscopeを用いた中枢時計のイメージング技術確立を目指してきた。また、UCLA、コロンビア大学、コーネル大学で、中枢時計研究のディスカッションや、国際交流を進めることができた。また、米国神経科学会、米国時間生物学会、アジア時間生物フォーラム、中国・日本時間生物交流会、サッポロ時間生物国際シンポジウム、その他国内の国際シンポジウムなどに参加・発表し、博士学生も含め、国際研究交流を行うことができた。
