Outline of Annual Research Achievements

本研究ではコロナ禍にも関わらず国際共同研究を展開することができた。分担者の河村はハンガリースポーツ科学大学との共同研究で心肺体力とDNAメチル化に基づく生物学的老化との関係を明らかにする研究で成果をあげることができた。高齢男性144名を対象に、心肺体力および生活習慣関連指標とDNAメチル化年齢との関係を検証した。その結果、年齢、飲酒および喫煙などの交絡因子で調整した後も、高い心肺体力が生物学的老化の遅延と関連することを明らかとなった。また、この研究では、心肺体力だけではなく、適切な代謝状態の維持、微量栄養素の十分な摂取、喫煙や過度な飲酒の回避、朝型の生活リズムなどの生活習慣がエピジェネティック時計の進行遅延と関連することが明らかとなった(Kawamura et al., Aging Cell, 2024)。次に、心肺体力に基づき繁殖を繰り返したラット系統を対象に、心肺体力と多臓器での生物学的老化との関連を検証した。その結果、高い心肺体力は生物学的老化の指標であるグローバルな平均メチル化レベル(Global Mean DNA Methylation)およびメチル化の混沌状態を表すDNAメチル化エントロピー(Mean DNA Methylation Entropy)の加齢変化を臓器特異的に遅らせることが示された。加えて、この研究では、海馬、心臓、ヒラメ筋および大腸の個別遺伝子のプロモーターメチル化レベルについて、一貫して心肺体力の影響を受ける7つの遺伝子と、各臓器別に最も影響を受けるトップヒット遺伝子をそれぞれ特定した。これらから、高い心肺体力は、生物学的老化の臓器特異的な遅延と関連することが示唆された(Kawamura et al., bioRxiv, 2024)。各組織からのエクソソームの抽出法についても比較検討を重ね、今後の研究に用いる手法について知見を得ることができた。
