Research Abstract |
転写酵素RNAポリメラーゼは、転写因子との相互作用で、ゲノムから転写対象遺伝子を選択し、転写因子の交換でゲノムの遺伝子発現パターンが変化する。ゲノム転写包括制御機構の解明を目標に、大腸菌の300種類転写因子の全てについて、制御支配下遺伝子群の同定と制御機能の解析を実施した。転写因子の制御標的については、本研究で開発したGenomic SELEX法で、純化転写因子が認識結合するDNA配列を決定することで同定した。分子生物学勃興期からのモデル生物である大腸菌でも、凡そ100種転写因子に関しては、機能未知であった。既知転写因子についても、制御標的のひとつまたは数個の遺伝子の解析に終わっていた。しかし、研究初年度で、凡そ半数の転写因子に関して、SELEX解析を実施し、標的の全体像を明らかにすることが出来た。一方、転写因子の特異抗体を作製し定量的免疫染色法で、大腸菌各種培養条件での細胞内濃度の決定を実施した。本年度、約100種の転写因子濃度を決定した。また転写因子の活性に影響を与える物質と環境要件に関しては、形質マイクロアレイ法(Phenotype microarray)で探索した。存在する転写因子の活性が、これら要素・要因で制御されていることが明らかになった。この戦略での研究を継続することで、ゲノム転写制御の全体像の解明を目指す。研究終了時には、ひとつの生物の、すべての転写因子の制御機能を解明を、世界に先駆けて成功させたい。
