献血を必要としない血小板輸血療法の戦略

Research Abstract

本課題では、ヒトES細胞、ips細胞をソースにした血小板輸血製剤の開発目的に、(1)特定分子の蛋白発現制御に基づく産生巨核球数の増多方法の開発、(2)新規のshear stress刺激に伴う血小板産生装置の開発、(3)血小板におけるメタロプロテアーゼ活性制御分子によるADAM10/ADAM17活性化・抑制の生理機構解明、の3つの課題の解明・解決を目指してきた。(1)に関しては、c-MYC遺伝子の発現パターンが正常な巨核球成熟およびその後の血小板産生を制御している事を見いだした(Takayama Net al., J Exp Med, 2010)。この発見をもとにcーMYCおよびBMI-1(ポリコーム遺伝子)を同時に造血前駆細胞に導入することで、巨核球が前駆細胞レベルで増殖継続可能である事を発見した(2011年12月の米国血液学会年次総会においてPlenaryセッションに選択)。これはc-MYC依存的にINK4A関連分子(p14,p19)の上昇に伴って発生した細胞老化現象をBMI-1が抑制した結果、細胞死を誘導せずにトロンボポイエチン(TPO)の細胞増殖作用に伴う巨核球のみの自己複製が成立したと推測される。一方、上記(2)の課題に対しては名古屋大学の福田敏男教授との共同研究により血小板放出デバイスを作製し、特許申請まで行ったが、既存の方法以上に血小板産生効率を高めることはできなかった。その結果、別のコンセプトによるデバイスの開発を新たに開始した。上記(3)の課題に対しては、外部製薬企業との共同研究から、培養血小板の不活性化阻害効果のあるADAM10/ADAM17阻害剤を開発し、その効果をiPS細胞から産生させた血小板を用いて検証し、そのin vivoでの機能を確認した(本薬剤が培養した血小板機能を保持できる事を確認)。最終的には血小板を冷蔵、冷凍保存させる事を目的として、薬の開発以外にも生理的な温度変化に伴う血小板の活性化、不活性化機構の解明を継続中である。
全体として、本研究費を使用したことで、課題の解明が進んだと考えられる。
