X線マイクロビームを用いた神経細胞回路作成技術の開発

Research Abstract

本研究課題"X線マイクロビームを用いた神経細胞回路作成技術の開発"では、X線マイクロビーム照射技術と半導体プロセス技術を細胞工学に取り入れて、神経培養細胞が任意のパターンに人工配列される神経回路(神経細胞チップ)の作成技術を開発した。
半導体プロセス技術によって培養基板上に細胞外マトリックスをパターン加工し、細胞は培養中において、パターン状にしたがって接着する。神経細胞チップ上に配置される培養細胞には、PC12細胞を用いた。通常、PC12細胞は、NGF成長因子の投与によって神経突起が成長する。また、強いX線や電磁波によっても、NGF成長因子の投与なしで神経突起を誘発することが報告されている。そこで、X線照射誘導によるPC12細胞の分化について実験を行った。PC12細胞のX線照射には、ビーム径10μmのX線マイクロビーム照射装置を用いて、細胞チップ上のPC12細胞を最大吸収線量20Gyまで照射し、インキュベータで10日間培養し、1日ごとに細胞の形態を観察した。その結果、X線によって分化誘導されるPC12細胞の効率は、吸収線量の増加とともに高くなり、15Gy付近で最大となって、その分化率は約14%であった。ただし、X線照射によって分化した細胞の神経突起は、NGFの投与にくらべて、小さいことが特徴である。
製作された細胞チップ上の神経細胞は、カルシウムイオン測定用の蛍光プローブ(Fura 2、Fluo 3)と刺激用微小電極を用いてその応答特性を調べた。通常培養条件のPC12細胞と比較して、大きな違いが見られないため、細胞チップ上のPC12細胞の培養に成功していることが確かめられた。
