| Research Abstract |
本年度はチオレドキシン(Trx)の血中滞留性を改善したヒト血清アルブミン(HSA)-Trx融合体(HSA-Trx)を作製し、ブレオマイシン(BLM)誘発肺線維症に対するその有用性評価を行った。
BLMをマウスに経気道投与することで、3日後のBALF中の炎症性細胞の浸潤及び14日後に肺の線維化が観察された。このBLMマウスにおいて、HSA-Trxの投与スケジュールの検討を行った結果、週1回の投与間隔で有意な線維化抑制効果が確認された。Trx単独との比較検討を行ったところ、HSA-Trxと同条件におけるTrxの単独投与では全く効果を示さなかったものの、Trx単独の2日に1回の腹腔内繰り返し投与によりHSA-Trxとほぼ同等の抑制効果を示した。本肺線維症モデルマウスにおける体内動態解析より,HSA-TrxはTrxと比較して血中滞留性の大幅な上昇と投与後2時間における肺組織移行性の有意な上昇が確認された。次に、本肺線維症に対するHSA-Trxの抑制メカニズムの解明を行った。酸化ストレス反応物である8-hydroxy-2'-deoxyguanosine及びnitrotyrosineの肺組織免疫染色並びに酸化ストレスによる組織傷害の指標である肺組織中malondiaidehydeを定量したところ、HSA-TrxはBLM誘発の酸化ストレスを有意に抑制した。加えて、この酸化ストレスにより惹起される炎症や浮腫に対する評価を行ったところ、HSA-Trxによる有意な抑制効果が観察された。以上の結果から,HSA-TrxはBLM誘発の酸化ストレスを抑制することで傷害や炎症を軽減し,最終的に線維化抑制効果を示す可能性が示唆された。
