環境化学物質のT細胞免疫応答攪乱による炎症性腸疾患増悪と大腸発がん促進機構の解明

Research Abstract

本研究は、環境化学物質曝露による炎症性腸疾患の発症・増悪のメカニズムの解明を目的とする。クローン病や潰瘍性大腸炎炎に代表される症性腸疾患は難治性であり、大腸癌を合併することの多い疾患である。その病因は不明であるが、腸菅粘膜局所におけるTh1、Th2、Th17による過剰免疫応答が関与していることが明らかになりつつある。平成21年度の本研究では、アゾキシメタン腹腔内投与により発がんを惹起し、デキストラン硫酸ナトリウムの自由飲水による大腸炎症を介した発がん促進系(AOM/DSS大腸発がん系)を用いて、強い酸化ストレス作用を有する環境化学物質トリブチル錫の大腸炎症増悪作用と発がん促進作用を明らかにした。本年度は、トリブチル錫曝露によるこの大腸発がん促進作用が、癌遺伝子や癌抑制遺伝子の突然変異を誘導することで発がんに関与することが明らかなactivation-induced cytidine deaminase(AID)の発現増強を介していることを示唆する結果を得た。研究代表者等は以前より、トリブチル錫は酸化ストレス作用を介してTh2細胞分化を促進することでTh2型免疫を増強することを示してきた。また、AID発現はTh2型サイトカインIL-4、IL-13により誘導されることが示されている。本研究より、トリブチル錫のTh2型免疫増強作用によるAID発現増強が間接的に発がんを促進している機構が推定された。
