光合成における翻訳のレドックス制御と環境応答の分子機構

Research Abstract

光化学系II反応中心の再生には、D1タンパク質など反応中心を構成するタンパク質の新規合成が必要である。申請者らは近年、強光ストレス下で発生する活性酸素によって、これらのタンパク質の合成が翻訳伸長過程で阻害されることを明らかにした。さらに、タンパク質合成阻害のメカニズムとして、翻訳伸長反応を担う翻訳因子EF-Gが酸化され、特定のCys残基間にジスルフィド結合を形成して失活することを明らかにした。
本年度の研究では、シアノバクテリアSynechocystis sp. PCC 6803の葉緑体型EF-G(Slr1463)に着目して、酸化の標的Cys105をSerに改変したEF-Gを発現する変異株を作製し、強光ストレス下でのタンパク質合成および光化学系IIに対する影響を調べた。改変型EF-Gを野生型EF-Gとともに共発現させた株では、全般的なタンパク質合成は影響を受けず、D1タンパク質の新規合成のみが著しく促進した。さらに、この変異株では光化学系IIの光阻害が緩和した。光化学系IIの光損傷の過程が影響を受けなかったことから、修復が促進していることが考えられる。すなわち、EF-Gの改変によってタンパク質合成系の酸化ストレス耐性が増大し、その結果、修復の酸化ストレス阻害が抑制され、光阻害が緩和したことが示唆される。
