MCP‐1を介した間葉系幹細胞による胃癌組織へのホーミング分子機構の解析

Research Abstract

我々は前年度まで,幹細胞マーカーの一つであるCD44およびCD133陽性細胞の局在を検討してきた,我々の検討によれば,胃癌組織において両者の陽性細胞は多数認められ,またCD44陽性,CD133陽性細胞が胃癌組織中に存在していることも確認できた.
こうした,事実は胃癌組織内に癌幹細胞と考えられる細胞が確かに存在していることを裏付けるものである.同時に我々は,胃癌の増殖や血管新生と深く関わっているCOX-2の発現レベルとCD44,CD133陽性細胞が相関していることも確認している.以前,我々はH.pylori感染MNU投与胃癌モデル(スナネズミモデル)を用いて,誘導された胃癌組織中にCD133陽性細胞が存在していることを確認している(Futagami S,et al.Celecoxib inhibits CD133-Positive cell migration viareduction of CCR2 in Helicobacter pylori-infected Mongolian gerbils.Digestion,2010,81,193-203)が,今回の知見は実験モデル同様に,ヒトにおいても,CD44+/CD133+細胞が胃癌の進展過程に関与している可能性を示すものと考えられる.
我々はこうした癌幹細胞が胃癌進展に重要な役割を演じていると考えており,この幹細胞のホーミングについても現在検討を続けている.我々の検討によれば,CD44+/CD133+細胞は種々のサイトカインIL-1β、VEGF,SDF等によって遊走が制御されているわけでないことがわかった.
Transwell membraneを用いたmigration assayを行ったところ,これらの中和抗体によっても必ずしも抑制されないことが判明した.
