C型肝炎ウイルス複製複合体に含まれる蛋白の同定とウイルス複製における役割の解析

Research Abstract

C型肝炎ウイルス(HCV)のゲノム複製を担う複製複合体(RC)はHCV非構造蛋白質と宿主因子から構成されると考えられるが、HCV複製調節に関わる宿主因子は十分に明らかにされていない。本研究では、プロテオミクス技術を利用してHCVRCを構成する宿主因子を同定し、その機能解析を行った。Membrane flotation assayによりHCVRCを含む膜分画を粗精製し、二次元蛍光ディファレンシャルゲル電気泳動と質量分析法によってHCVRC分画に含まれる宿主因子を同定した。二次元電気泳動にて約1300蛋白が検出され、45蛋白(3.5%)が、高HCV複製期に1.5倍以上増加していた。その中には二種類の分子シャペロンT-complex polypeptide 1 ring complex subunit ε(CCT5)及びHeat shock cognate protein 70が含まれていた。ウエスタンブロット解析から、両蛋白は細胞の増殖相によって発現レベルに差は認められないものの、高HCV複製細胞の膜分画で明らかな量的増加を確認した。また、エピトープタグ免疫沈降法により、CCT5がHCV NS5B蛋白と特異的に結合することを見出した。さらに、CCT5 siRNAによってHCVRNA複製レベルが低下すること、また薬剤処理によってT-complex polypeptide 1 ring complexを細胞内で誘導したところHCV複製が亢進することを見出した。CCT5を含むT-complex polypeptide 1 ring complexは、本来細胞質でシャペロンとして作用し、蛋白質の機能的な立体構造形成に寄与する。今回同定された分子シャペロンがNS5B蛋白との結合を介してHCV RCへ取り込まれRNA複製調節に関与する可能性が示唆された。
