免疫難病モデルマウスを用いた生物学的製剤の分子作用機序の解明

Research Abstract

クローン病モデルマウス:22年度までの研究において、クローン病モデルマウスにおけるTNFα阻害療法とIL-6阻害療法の比較を行い、その治療効果および作用機序の違いについて炎症性腸疾患専門誌に報告した。結果であるが、TNF阻害抗体とIL-6R阻害抗体の両者が病原性ヘルパーT細胞の増殖を阻害するという類似の作用機序をもつことが明らかになった。一方、TNFα阻害作用をもつ「おとり型TNF受容体」(TNFR-Fc)はTNF阻害抗体と異なり全く無効であった。現在、TNF阻害作用をもつ2つの製剤がなぜ全く違う作用をもつのかに関してさらなる検討を加えており、さらに、新たに関節リウマチ治療薬として認可された生物学的製剤(CTLA4-Ig : abatacept)のクローン病モデルマウスに対する作用についても追加検討している。
顕微鏡的多発血管炎(MPA)モデルマウス:22年度においてMPO欠損マウスより調整したリンパ球を採取し、リンパ球欠損マウス(rag2欠損マウス)へとトランスファーすることで疾患誘導を試みた。血管炎の特徴であるMPO-ANCA(抗好中球細胞質抗体)の測定を行い、さらに腎病変(急速進行性糸球体腎炎)や肺病変などの全身血管炎症状を示唆する所見を生化学的および組織学的に検討したが、これまでのところ想定した著明な病的変化を検出するに至っていない。現在、オプションとして想定していた別手法による疾患誘導プロトコールを試みており、今後、これまでの手法と比較しながら上記の血管炎所見について検索を行う予定である。
