薬剤耐性胃癌細胞株の樹立とそのタンパク分子プロファイル

Research Abstract

癌細胞の薬剤耐性のメカニズムを解明することは癌化学療法において重要である。根治手術後の補助化学療法の進歩に伴い、手術単独の治療よりも長期無再発・長期生存例が得られるようになってきた。一方、術後補助化学療法にも関わらず30-40%の再発が見られることも事実であり、そのような化学療法の効果が見られなかった症例への対応が急務である。
本研究では癌細胞がどのようにして薬剤耐性を獲得するのかの分子生物学的機構を明らかにするために胃癌細胞株MKN45にCDDPを添加し、薬剤存在下で生存しえたコロニー由来の細胞を増殖させ耐性細胞株として樹立した。この耐性株を親株とともに再びCDDP下での反応を比較するとDNA傷害に関わる経路の反応が破たんしていることが判明した。耐性株では高度の細胞ストレスに暴露された場合、アポトーシスや細胞周期の停止などの適切な状態に進むことができず増殖が停止しないことになる。逆相タンパクライセートアレイを用いたCDDPへの反応性の実験でも同様の結果が得られた。
