| Research Abstract |
1. 血中遊離癌細胞(ITC)の検出とITCの遺伝子発現解析:
血液中の循環癌細胞を直接測定・解析するシステムであるCellSearch^<TM>を用い、胃癌播種性骨髄癌症5症例の血液中からサイトケラチン陽性細胞(=癌細胞)を検出し、回収した。回収した細胞からRNAを抽出し、gene tipを用いて遺伝子発現のprofilingを行った。これらのITCで普遍的に強発現あるいは発現が低下しており、DIC (disseminated intravascular coagulation)などの病態に関与していると思われる分子の同定作業を現在行っている。
2. 原発巣、転移巣における癌細胞の性質:
1で得られた結果から、主な遺伝子をpick upし、原発巣、転移巣、末梢血液に存在する癌細胞で発現のパターンが変化しているのかどうか、免疫組織染色、Western blotting、RT-PCRなどで確認する予定であったが、まだ、遺伝子のpick up中である。
3. xenograftの作製:
CellSearchで分離・回収されたヒト胃癌細胞(ITC)を免疫不全マウスに接種し、2症例において胃癌播種性骨髄癌症モデルマウスが作製された。
4. 癌時幹細胞の同定:
1症例において、Xenograftの作製過程で腫瘍再現能が確認され、さらに二次移植で自己複製能が確認されたことにより、患者末梢血から得られた癌細胞中に癌幹細胞あるいは類似の細胞が存在したことが強く示唆された。
