| Research Abstract |
,1.血中遊離癌細胞(ITC)の検出とITCの遺伝子発現解析:
血液中の循環癌細胞を直接測定・解析するシステムであるCellSearch^<TM>を用い、胃癌播種性骨髄癌症5症例の血液中から癌細胞を回収した。回収した細胞からRNAを抽出し、gene tipを用いて遺伝子発現のprofilingを行った。これらのITCで普遍的に強発現あるいは発現が低下しており、本症の病態に関与していると思われる候補遺伝子を同定した。
2.原発巣、転移巣における癌細胞の性質
1で得られた候補遺伝子について、原発巣、転移巣、末梢血液に存在する癌細胞で発現のパターンが変化しているのかどうか、免疫組織染色でスクリーニングしたが、劇的にパターンが変化する分子の同定には至らず、解析を続けている。
3.xenograftの作製:
CellSearchで分離・回収されたヒト胃癌細胞(ITC)を免疫不全マウスに接種し、2症例において胃癌播種性骨髄癌症モデルマウスが作製された。
4.癌幹細胞の同定:
1症例において、Xenograftの作製過程で腫瘍再現能が確認され、二次移植で自己複製能が確認されたことにより、患者末梢血から得られた癌細胞中に癌幹細胞が存在したことが示唆され、解析を進めたが、この腫瘍は本来の胃癌とは異なり、virusにより発生した腫瘍であることが判明した。
5.治療への応用:
胃癌播種性骨髄癌症のうち2例のITCがHER2を発現していることが判明し、ハーセプチンを用いた治療を施行し、良好な治療効果を得た(論文投稿中)。
