新規関節形成マスター遺伝子SOX11を軸とした関節形成分子ネットワークの解明

Research Abstract

SOX11の発現パターンについてマウス発生過程において経時的に解析した。まず胎生11.5-12.5日のマウス胚においてwhole mount in situ hybridizationを行ったところ、未分化間葉系細胞の凝集部位もしくは未分化軟骨細胞の部位に局所的な強い発現がみられた。続けて組織切片においてin situ hybridizationを行ったところ、成長とともにSOX11は関節形成部分に限局して発現するが、関節が形成された後は成熟するにかけて発現は漸減していくことが分かった。
次にSOX11の転写標的として関節マーカー遺伝子、軟骨基質遺伝子、軟骨内骨化関連遺伝子、軟骨変性関連遺伝子などを広く検討した。SOX11はGDF5、WNT9Aなどの関節マーカー遺伝子だけでなく、2型コラーゲンやアグリカンなどの軟骨全般に発現する基質タンパクを広く誘導することが分かり、それぞれの遺伝子について転写解析を行って応答領域を同定することができた。反対に軟骨内骨化関連遺伝子や軟骨変性関連遺伝子については発現を抑制する傾向があることが分かったが、その詳細なメカニズムについては解明できなかった。軟骨基質誘導についてSOX9など既知の分子との協調性があるかどうか検証したが、相加的な効果にとどまり直接の関連はないことが分かった。またルブリシンなど関節軟骨の成熟とともに発現が増す遺伝子についても検証したが、SOX11はこれらの誘導には深い関係はなく、関節軟骨の発生過程については関与していても、その維持については関与していないと考えられる。
