子宮内膜症病態メカニズムに関与するリポキシゲナーゼ経路関連因子の解析

Research Abstract

本研究では、リポキシゲナーゼ(LOX)経路および関連する脂質mediatorの働きに着目しながら、細胞分子生物学的な解析手法により、子宮内膜症の発症・進展・慢性化機序の一端を明らかにすることを目的とする。本年度は、子宮内膜症の病態メカニズムにおけるin vivoでのLOXの役割を明らかにするために、前年に引き続き、マウスを用いたin vivo異所性内膜モデルの更なる開発を主に行った。重度免疫不全マウスに対して、分散したヒト子宮内膜細胞を尾静脈より投与し、血行性に異所性生着する内膜症モデルマウスの作成を試みた。その解析対象臓器は、子宮、卵巣、肺、腹膜、消化管などとして、ヒト子宮内膜を構成する腺上皮、間質、血管内皮などの各細胞を特異的に認識する抗体を用いた免疫組織化学、ならびにヒトDNAを特異的に認識するプライマーを用いたreal-time PCRによりヒト組織の定量化を行った。一方、LOX関連遺伝子の導入を念頭において、まずluciferase(LUC)ならびにgreen fluorescent protein(GFP)などの標識遺伝子を組み込んだレンチウイルスの作成を行った。必要に応じて、このレンチウイルスを用いて移植内膜細胞をLUCおよびGFPで標識した。また、本モデルに同時に腹腔内に同じ分散内膜細胞を投与し、LOXが関与し得ると考えられる腹腔内炎症を惹起することで、異所性内膜の発生や進展にどのような影響が及ぶかについても検討した。これまでの結果として、移植内膜細胞は主に肺に集積しその後徐々に減少したが、移植後20週以降でも少なくとも肺実質にはヒト内膜由来の細胞集落を免疫組織化学で確認し得た。さらに、real-time PCRでもヒト由来DNAがマウス肺に認められた。以上より、異所性内膜症、特に肺内膜症のモデルになり得ると考えられた。
