Research Abstract

シグレックは免疫グロブリンスーパーファミリーに属し、シアル酸を含む糖鎖を認識するタンパク質の総称である。その一種であるシグレック2(CD22)は、α2-6シアル酸を特異的に認識し、抗体産生細胞の前駆細胞であるBリンパ球に発現する。本年度は、シグレック2とリガンド糖鎖の親和性を解析するための分子プローブの開発を目的として研究を行った。これまでの研究でシグレック2に一定の親和性を有することが知られている9-biphenyl-NeuGcα2-6Galをモチーフにし、還元末端にアルキンを導入後、末端にビオチンを有するアジド体をクリックケミストリーによって縮合させ、目的とする分子プローブを合成することに成功した。本プローブを用いることでELISA法でのスクリーニングが可能になり、本プローブを用いて系統的な類縁体の設計・合成とシグレック2に対する親和性の評価を進めている。
一方、MBPはマンノース結合型タンパク質あるいはマンノース(マンナン)結合レクチンとも呼ばれるC型動物レクチンである。MBPはある種のガン細胞に対して細胞障害作用を持つことが示されており、リガンド糖鎖としてLe^aエピトープの繰り返し構造が提案されている。本研究では、MBPのリガンド構造要求性の解明を目的として、繰り返し単位1～5までのLe^aタンデム構造の構築を目指している。本年度は繰り返し単位1～3のLe^aタンデム構造を構築したが、得られた糖鎖はMBPと有意な親和性を示さなかった。
