糖尿病大規模コホート研究におけるミトコンドリアDNA解析

Research Abstract

〈研究目的〉
九州大学の久山町コホート研究グループは、近年、年齢、性別等をマッチングさせた3000人以上のゲノムサンプルの収集を終え、長期前向きのコホートスタディーを新たに開始している(例数はさらに増加中)。特筆すべきはこれら解析対象者全員について、生活習慣情報や一般検査情報に加え、糖負荷試験による精密な耐糖能情報が得られていることである。申請者は共同研究として、この大集団のミトコンドリアDNA配列を解析することで、これまで誰も明確に示すことが出来なかったミトコンドリアDNA多型と糖尿病発症リスクとの関連にはっきりとした結論を出すことを目的としている。
〈研究成果〉
ミトコンドリアDNAの転写・複製の制御部位であるD-loop領域の約1kbpと電子伝達系複合体Iサブユニット6をコードする約1kbpをターゲットとして、上記3000人以上の全配列を決定した。現在この配列データを基に、疾患との連鎖する多型について検索中である。それに加え、22年度は一定の糖尿病診断基準を満たした5000人の糖尿病患者の血液生化学検査値とトータルゲノムを収集した。
