乾燥無代謝休眠覚醒過程に特異的な生体分子修復系の分子背景の解明

Research Abstract

吸水過程のネムリユスリカを用いてcDNAライブラリーを作製し、その配列をデータベース化した。具体的には、乾燥幼虫(吸水0時間)、吸水1時間及び吸水24時間の幼虫を用いた。同時に、発現産物のカタログ化を行う目的で、乾燥過程及び吸水過程などの様々な処理を施した幼虫から抽出したRNAを混合し、平均化ライブラリーを作製した。併せて4つのライブラリーから、各10,000個のコロニーを単離し、5'及び3'末端の塩基配列を決定した。得られたデータセットを、作成済みの乾燥過程ESTデータベースに加えて、再度クラスタリングを行ったところ、70,860クローンのESTが、24,183個のクラスターに集約された。得られたクラスターには、発現頻度が低いと思われる遺伝子群である転写因子やシグナル因子も多く含まれていた。
今回作製したデータベースには、DNA修復遺伝子群も含まれていた。例として、塩基除去修復に関連するRad23と二本鎖切断修復に関与するRad51があげられる。これらの遣伝子の発現をReal-Time PCRで定量したところ、Rad23は吸水過程のみならず乾燥過程でも発現が上昇していた。一方、Rad51は吸水過程で顕著に発現が上昇していた。このことから、乾燥過程と吸水過程では、種類の異なるDNA障害が生じており、この障害を修復する機構が作動していることが明らかとなった。
この結果は、22年度内に論文化し、詳細を報告予定である。
