乾燥無代謝休眠覚醒過程に特異的な生体分子修復系の分子背景の解明

Research Abstract

乾燥無代謝の覚醒過程に起こる修復系の分子背景の解明を目的に、乾燥及び再水和過程の網羅的な遺伝子発現解析を行うことにした。
昨年度作成したネムリユスリカESTデータベースから独立したクラスターを抽出するために、Blast-xに基づく再クラスタリングを施した。カスタムマイクロアレイを作製するために、得られたクラスターデータを元に、独立した遺伝子をコードするプローブを選抜した。最終的に、16,652個のプローブを選抜し、アジレント社製の4×44kフォーマットのアレイを作製した。その結果、乾燥前に比べて、乾燥後24時間後または48時間後に10倍以上発現上昇する遺伝子は少なくとも825個存在し、1/10以下に発現が減少する遺伝子は少なくとも171個存在することが明らかとなった。一方、乾燥したユスリカに比べて、再水和後24時間後に10倍以上発現が上昇する遺伝子は少なくとも803個存在し、1/10以下に発現が減少する遺伝子は少なくとも217個存在することが明らかとなった。これらの情報に基づき、発現クラスター解析を行っている。
再水和過程でのDNA修復糸の検討も進めた。吸水過程は著しい酸化ストレスに曝される過程であり、その結果DNAが損傷受けていた。この損傷に対する修復系や活性酸素の除去系が、再水和に伴って作動することが、乾燥無代謝休眠からの健全な覚醒に重要であることが明らかとなった。同時に、この過程には、HSPも関与していることも明らかになった。
以上の本年度までの結果を論文にまとめ、PLoS OneやJBCなどのジャーナルに報告した。
