マウス及びヒトの腸管粘膜固有層に存在する抗原提示細胞群の機能解析

Research Abstract

本研究課題は、マウス及びヒトの腸管粘膜固有層に存在する抗原提示細胞群の機能を解析することを目的としている。
マウスの小腸にはTLR5を発現しているCD11c^<high>CD11b^<high>DCが存在しており、粘膜固有層におけるIgA産生形質細胞の分化や、抗原特異的なCD4_+ヘルパーT_H1細胞とT_H-17細胞の分化をTLRの刺激依存的に誘導出来ることを明らかにした。粘膜固有層にはCD11c^<high>CD11b^<high>DCの他に、CD11c^<low>CD11b^<high>DCが存在している。この細胞は、MHC classII^<high>, CD103+DCで、補助刺激分子のCD80やCD86を発現していることを明らかにした。CD11c^<low>CD11b^<high>DCは、TLR3やTLR9を発現しており、それらのリガンドに反応してinterleukin(IL)-6やIL-12などの炎症性サイトカインを産生した。そして、CD11c^<low>CD11b^<high>DCは、naive CD4 T細胞に働きかけ、TLR刺激依存的にTh1細胞を誘導することをあきらかにした。
ヒト腸管粘膜固有層から収量よくかつviabilityの高い抗原提示細胞を回収するために、酵素の濃度や処理時間を検討し、ヒトの腸管における最適な単離法の確立を行った。
