| Research Abstract |
We used helper-dependent adenoviral vectors to knock-out KU80, LIG1 and LIG3 genes by homologous recombination in human induced pluripotent cells (hPSCs). Unlike heterozygous mutants of LIG1 and LIG3, which showed a tendency for sensitivities to a reagent causing DNA single-strand break, the KU80 heterozygous hiPSCs did not show any obvious phenotypes. Furthermore, increase in gene targeting efficiency was not observed in these heterozygous mutants.
