Research Abstract |
We found the insulinotropic effects of GLP-1, sulfonylureas and free fatty acid (FFA) are enhanced in mouse pancreatic islets lacking leptin signaling. GLP-1-induced intracellular cAMP-CREB pathway was enhanced in those β-cells. Furthermore, the detrimental effect of chronic FFA treatment on insulin secretion, or "lipotoxicity", was augmented in islets lacking leptin signaling. These results indicate leptin signaling modulates nutritional or therapeutic insulin secretion in pancreatic β-cell.
