Outline of Final Research Achievements |
We recently found that almost any enzyme can become biosensor component by moderately destabilized through random mutations. In this study, we have developed the technology to improve enzymes, biosynthetic pathways, and mocrobial hosts using biosensors. Also we have also succeeded in fusing multiple enzymes in the ways where fusion partners are mutual regulating to each other. Furthermore, we have demonstrated that temporary becoming ligand-induced folder allows protein biosensors to rapidly access to the novel sensor functions.

Academic Significance and Societal Importance of the Research Achievements |
本研究は,「Ligand-induced folding」という現象が,ランダム変異の蓄積によって,酵素を含むあらゆるタンパク質に頻出する現象であることを改めて示すとともに,これが代謝工学や宿主工学,そして酵素工学のための有効なスクリーニングツールを提供できることを明確に示すことができた.また,自然界で起きているタンパク質の機能進化において,これまで見過ごされてきた要素を提示するとともに,より複雑なタンパク質の協働機能の設計学,Protein Computingやタンパク質機能への制御機構の埋め込み技術としての新しい可能性を示すことができた.
