Outline of Annual Research Achievements

2022年度は以下の2つの項目に関して次のような進捗が得られた。
1. ラベルフリー代謝イメージングによるマウス肝臓の代謝構造可視化: 本課題で開発した「光干渉断層顕微鏡 (OCM; optical coherence microscopy)を基盤としたラベルフリー代謝イメージング手法(LIV; logarithmic intensity variance法)」を用いることで、ex vivo マウス肝臓における代謝の活発な領域の三次元可視化に成功した。この研究では、摘出したマウス肝臓を経時的に三次元計測し、LIV手法で処理することで代謝構造の可視化を行った。マウス肝摘出直後には血管構造の直下に高い代謝が見られた。この高代謝は12時間程度で一旦消失し、その後、再度高い代謝(もしくは何らかの組織・細胞内運動)が発生することが確認された。さらに、この再出現した活動は摘出後36時間程度で消失することが確認された。
2. Iv vivo zebrafish の内部構造可視化: 本課題で開発された multi-contrast OCM、および、高分解能OCM装置を用いて、in vivo および postmortem の zebrafish の全身をイメージングすることに成功した。この研究により、Zebrafish の筋組織が高複屈折領域として、また、脊索が低複屈折領域として描出されることが確認された。また、水晶体においては放射上の繊維束構造が可視化されることが確認された。これらの結果により、本課題で開発している OCM 装置が zebrafish model を用いた医学・生物学研究にも利用可能であることが示唆された。

Current Status of Research Progress

1: Research has progressed more than it was originally planned.

Reason

当初計画では postmortem zebrafish および ex vivo マウス臓器を対象としたラベルフリーイメージングを実現する予定であった。しかしながら、研究初期におけるトライアルによって、zebrafish では in vivo で計測が可能であることが示唆された。そのため、当初計画を前倒しして、in vivo でのイメージングを行い、その結果を Biomedical Optics Express 誌に掲載した(10.1364/BOE.455876)。
また、本研究では、本年度は低分解能のマルチコントラストOCM装置を中心として研究を進める予定であった。しかし、プロジェクト開始後に、新たに高分解能OCM開発を開始し、当初予定よりも早い段階で実用的な画像の取得に成功した。(この装置は上記の zebrafish 研究でも利用されている。)
以上の理由により、本課題は当初の計画以上に進展している、と考えられる。

Strategy for Future Research Activity

上記の研究の中で、「多重散乱光によるノイズ」が深部のイメージングにおいて大きな問題となる(ノイズ源となる)ことがわかってきた。そこで、2022年度は、新たに多重散乱光除去手法の開発を進める。この手法は、新たにOCM装置に組みいれる可変焦点レンズ、それを制御することで実現する「focus modulation 計測」、および、当初計画でも開発を予定していた holographic refocusing を組み合わせることで多重散乱光を除去する手法である。この開発により、1-2 mm 深さでの高精度なマルチコントラスト画像の撮影画可能になると考えられる。この画像化深度は、上記の zebrafish であれば、その full depth をイメージング可能な画像化深度である。
