Outline of Annual Research Achievements |
We analyzed hemangioblast specification by ETV2 and NPAS4L (hemangioblast regulators),and by SNAI2 and ZEB2 (EMT regulators). We showed that ETV2 gene was deleted in all 363 avian genomes, and that NPAS4L functioned as key chicken hemangioblast specifier. Mammalian ETV2 induced vessel, but not blood differentiation in chicken;as with gecko ETV2 (gecko contains both both NPAS4L and ETV2 genes). We tested whether hemangioblast specification was affected by EMT regulators. Endogenous SNAI2 was expressed in the streak and nascent mesoderm cells, without overlapping with NPAS4L, as with ZEB2. Overexpression of chicken SNAI2 did not affect normal expression of SCL/TAL1, LMO2 or HAND2, suggesting that hemangioblast differentiation is regulated separately from epithelial-mesenchymal morphogenesis.
