ミクログリアによる感覚モダリティーの制御と精神病態への寄与

Outline of Annual Research Achievements

本年度はミクログリアによる異種感覚の可塑性への寄与を明らかにし、その精神疾患病態への関わりについて検証した。
まず、高次視覚野であるV2Lにそれぞれの第一次感覚野から投射があることを示し、V2Lにおける神経細胞の機能応答を検証したところ、通常ではヒゲ刺激によってV2Lの応答は抑制されるが、視覚遮断によってその活動頻度が増強することがわかった。詳細な検討を行ったところ、ミクログリアは視覚遮断に伴い、興奮性神経細胞周囲に接触し、抑制性シナプスを剥がすことによってその興奮性神経細胞の興奮性を獲得することに寄与することがわかった。さらにこの獲得した興奮性はヒゲの感覚識別に寄与することも明らかにした。これは先天的視覚遮断によって起こる現象である。では後天的視覚遮断ではどうなるのであろうか。ヒトでは極度の低視力もしくは視覚喪失に伴って幻覚が生じることが知られている(シャルルボネ症候群)。マウスでこれを検証するために成熟マウスで視覚遮断を行い、神経細胞の活動を検証したところ、神経細胞集団の活動の増加、ミクログリアの活性化それに伴うアストロサイトの活動増強が観察された。これらの神経活動に対する寄与を現在検証するとともに責任分子の同定をそれぞれの細胞のRNAシークエンスを用いて行っている。さらにこのミクログリアの精神疾患での変化を形態変化で検証したところ、突起数の減少を認めた。これは大脳皮質全般で起こる変化であり、トランスクリプトーム情報との統合を行っている。
