Outline of Annual Research Achievements

本研究課題では、ゲノム編集マウス作製・解析プラットフォームを完成させて応用し、哺乳類の精子・受精のバイオロジーを究める研究を同時に進めている。
1)ゲノム編集マウス作製・解析プラットフォーム:23年度も、Lipid NanoParticle(LNP)を用いたmRNA導入によるゲノム編集技術開発に取り組んだ。LNPは透明帯を除去することなく受精卵にmRNAを導入できることを利用して、Floxマウスの受精卵にCre mRNAを導入し、ほぼ100%の割合でFlox領域を抜き取ることに成功した。ウイルスベクターや顕微操作を必要としない簡便な受精卵mRNA注入法を確立した。またCas9を全身で発現する遺伝子改変マウスを用いてゲノム編集マウス作製を試みたが、Cas9発現量が少ないためか、実用化には至らなかった。一方で、アデノ随伴ウイルスベクター(AAV)を用いた遺伝子改変マウス作製に取り組み、特定のセロタイプを有するAAVが受精卵への遺伝子導入に適していることを見出した。
2)哺乳類の生殖生物学:精巣特異的に発現する104遺伝子について、KOマウスを作製して交配試験を実施した。その結果、2系統で胚性致死を示し、残る102系統のうち、60系統では妊孕性に問題がなく、8系統で妊孕性が低下、34系統で雄性不妊が認められた。雄性不妊系統については、さらに解析を進め、①CCDC183がないと精子鞭毛形成の際の細胞質陥入が上手くいかないこと、②TEX46がないと精子頭部の形態異常を起こし透明帯を通過できずに雄性不妊となること、③NELL2と共同して精巣上体の分化誘導に働くNICOの同定、などを論文報告した。また国際共同研究により、ZP2の切断による透明帯硬化メカニズムや精子CaチャネルであるCATSPER集合体形成メカニズムに関する論文を報告した。

Reason

2023年度は当初予定を上回る104遺伝子のKOマウスを作製し、表現型解析を実施することができた。代表者が責任著者としてだけでなく、高インパクトを有する国際共著論文も多数報告できており、当初の計画以上と判断した。

Strategy for Future Research Activity

研究は順調に進展しており、当初予定通り推進することができると考えている。KOマウス作製による生殖関連遺伝子の機能解析は、競争が激しくなってきているが、国際連携を高めることで、競争力を維持したい。
