Outline of Annual Research Achievements

This study aims to develop suitable metal-mediated base pairs that can be enzymatically incorporated into the DNA. During my last year’s research, I have made several progresses as listed below:

1.Designed and synthesized several novel nucleobases based on the criteria concluded from the previous research and obtained oligonucleotides containing one or more such base pairs. Apart from the ImOC, which was reported previously, the TazC is a novel candidate.
2.Thermal melting analysis of DNA duplexes containing artificial nucleobases were conducted in the presence and absence of various metal ions, in which the TazC was found to form stable TazC-Cu(II)-TazC base pair.
3.ESI-TOF MS measurements were conducted on the TazC base pair, and the formation of the Cu(II)-mediated TazC base pair was confirmed.
