分子間協働から創出される細胞容積調節能を利用した虚血性壊死からの救済策の開発

虚血性疾患は、世界で最も大きな死因の一つである。虚血性傷害は、壊死に陥った組織を回復させることは極めて困難であるため、壊死を未然に抑えるための治療法が求められている。虚血誘発性細胞死は、虚血後に急速な細胞膨大が続き、最終的に壊死となる。そのため壊死を防ぐためには、細胞膨大させる機構を知る必要性が高まった。細胞膨大に働くイオンチャネルは、最近の研究結果によりTransient Receptor Potential 7 (TRPM7)が分子実体であることが分かり、より詳細な解析を本研究で行った。
今回の研究では、細胞容積調節機構に膨らみのセンサーであるカチオンチャネルのTRPM7タンパクと、元の細胞の大きさに戻すアニオンチャネルの容積感受性外向き整流性クロライドチャネル(VSOR)タンパクの2つのタンパク質が何らかの役割を担うだろうという、世界で議論になっていた疑問に対するを証明をした。ここでは、細胞の膨張を抑えるシステムの中でどのような関係で働いているのかについて、二つのタンパク質(TRPM7とVSOR)が協調することで、膨らんだ細胞が元に戻るしくみを明らかにした。ひとつには、細胞膨大時にTRPM7のC末端とVSORのコア分子であるLRRC8Aが物理的に相互作用すること、二つにはTRPM7から入ってきたカルシウムイオンののちにVSORが機能連関で活性化することを二種類のイオンチャネルの記録を同時に経時的に行うことで証明した。
これらの結果は日本生理学会などで学会発表を行うとともにプレスリリースで世間に広く情報を公開した。
