Outline of Annual Research Achievements

Cancer-specific monoclonal antibody (CasMab) 法は、がん特異的な反応性を示す抗体を樹立する方法として我々が独自に開発した技術である。我々はこれまで、このCasMab法を用い、細胞表面に発現するI型膜タンパク質の腫瘍特異性を特徴づける表現型を識別する抗体を戦略的に樹立してきた。
本研究課題では、がん細胞、正常細胞の両方に発現しているタンパク質を標的とし、既に抗体医薬が開発されているがより優れた抗体が求められるもの、または、抗体医薬が存在しない新規のものを標的分子として、がん特異的抗体の樹立を目指した。そして、樹立したがん特異的抗体を利用し、腫瘍特異性を担う原因の実態を解明し、腫瘍型標的分子のがん細胞形質への関与を解析する計画であった。また、がん特異的抗体の抗体医薬開発の可能性を、遺伝子改変技術を用いて検証する計画も遂行した。
研究計画に基づいて、研究期間を通して、継続して標的分子に対する抗体ライブラリーの作製を実施した。種々の標的分子に対して、100クローンを超えてハイブリドーマの樹立を行なった。樹立した抗体について、順次、種々のアプリケーション(FACS, WB, IHCなど)への有用性を検証した。標的分子に対して標準となる候補抗体1点以上について、また、いずれかのアプリケーションでCasMab傾向を示した候補抗体について、エピトープ探索実験を遂行した。解析に進んだ候補抗体については、抗体遺伝子を取得し、必要に応じて遺伝子組み換え等により抗体活性の検証を行なった。
最終年度であるR5年度に、HER2に対する新規CasMab抗体について、抗体の樹立からエピトープ解析までの結果を論文として発表することが出来た。また、共同研究による立体構造の解析結果から、このHER2に対するがん特異的抗体の腫瘍特異性を担う原因が示唆された。
