| Outline of Annual Research Achievements |
多種類の自己免疫性水疱症のうち、抗p200類天疱瘡については、私どもが、ラミニンγ1が自己抗原の一つであることを示したが、現在まで真の自己抗原とその病原性は解明されていない。
3年間の本研究では、3年間継続して、多くの自己免疫性水疱症症例の診断検査を施行し、約30名の新たな抗ラミニンγ1類天疱瘡症例を渉猟した。また、3年間に自己免疫性水疱症に関連した約70報の英文論文を発表したが、その内の8論文は、抗p200類天疱瘡に関する論文であった。本研究の1年目には、主に、ラミニンγ１について、そのリコンビナント蛋白とCRISPR-Cas9法によるノックアウトHaCaT細胞を作製した。2年目は、ラミニンγ１ノックアウト細胞を用いた抗p200類天疱瘡の研究を進め、Osaka City Med Jに論文発表した。
本研究の当初の目的は、抗p200類天疱瘡における抗ラミニンガンマ１自己抗体の解析であったが、ドイツのリューベック大学の研究者との検討で、ラミニンベータ４も自己抗原であること見出した。そのため、最終年度の3年目は、ラミニンガンマ１とラミニンベータ４の両方について検討することにした。まず、ファージディスプレイ法に必要な機材と試薬を準備し、抗p200類天疱瘡患者末梢血から単離したB細胞より抽出したRNAのcDNAを用いてファージライブラリを作成した。同時に、ラミニンベータ４のリコンビナント蛋白を作製した。また疾患モデルの準備として、患者血清より精製したIgG抗体を用いたex vivoヒト皮膚切片モデルを確立した。
今後、リコンビナント蛋白を用いたパンニングを繰り返して2種の自己抗原の病原性モノクローナル抗体を得る。その後、その病原性モノクローナル抗体を用いて、ex vivoヒト皮膚切片実験や新生マウス動物実験などの疾患モデル実験研究を行う予定である。
