Outline of Annual Research Achievements |
The purpose of the proposed research was to establish deep ultraviolet (DUV) fluorescence microscope for 3-dimensional (3D) brain imaging specifically towards mesoscopic quantification of the anatomical structures of interest. Secondary goal included developing or adapting open microscopy and open data toolkits for 3D image acquisition and analysis.
The first phase (FY2021) of the development of fluorescence microscope for whole brain imaging was successfully implemented. This included extending the fluorescence microscope towards automated serial block-face imaging of the whole brain. Image acquisition and analysis pipeline was developed using custom developed open source based software using Python as the programming language. A graphical user interface was also developed for image acquisition (FY2022). A technical publication in a peer reviewed journal was published in FY 2023. Adapting existing open source tools for our microscopy system was also carried out using Micro-manager, which would provide ease of adoption and usage in the community (FY 2023). In FY 2023, a conference presentation on open microscopy for DUV microscope was done. Consolidation of this work for journal publication is under consideration.
