Outline of Annual Research Achievements |
Isolation, purification, and quantification: -Momilactones A (MA) and B (MB) were completely isolated and purified from rice husks, of which, 100 mg MA and 200 mg MB from 20 kg rice husk. On the other hand, we completely isolated p-coumaric acid and tricin from rice husk. The purification is in progress. In addition, we preliminarily quantified MA, MB, p-coumaric acid and tricin in some Japanese rice varieties (brown rice). A new method for optimizing the detection of momilactones and other active compounds was developed. Screening of anticancer activity (cytotoxicity): -We completely investigated the cytotoxicity of MA and MB on blood cancer cell lines including Meg-01, K562, HL60, Mino, U266, KMS-11 by MTT assay. We completely achieved IC50 value for the cytotoxicity of MA and MB on both tested normal cell and cancer cell lines.

2: Research has progressed on the whole more than it was originally planned.
Reason
- We had a good preparation for extracting and isolating target compounds from rice husks, therefore, I could get the desired amount of momilactones and relevant compounds; - We had a good collaboration with Hematology Lab led by Prof. Dr. Takami Akiyoshi at Aichi Medical University, therefore, the screening cytotoxicity of momilactones on blood cancer cell lines could be done successfully; - We have not carried out cytotoxicity on lung and gastric cancer cell lines because of the delay in ordering cell lines and chemicals caused by COVID-19.

Strategy for Future Research Activity |
- To continue isolating and purifying bioactive compounds and screening the cytotoxicity on other cancer cell lines; - To determine anticancer mechanism of isolated compounds, especially, momilactones A and B; - To evaluate the effect of in vitro digestion on accessibility, availability, and activity of momilactones A and B.

Causes of Carryover |
The incurring amount will be used for the next steps of the project which mainly focus on the experiments implementation about the anticancer mechanism and the effect of the digestion system on bioactivity of momilactones and active compounds from rice.
