Outline of Annual Research Achievements

Synthesis protocol of silver nanowires (AgNWs) was completed and the purification and length control protocols of AgNWs were established. Amyloid nanofibrils (ANFs) were influential in the morphological modification of spray-dried supraparticles (SPs) containing silica nanoparticles (SiNPs; 10 and 100 nm in diameter) and AgNWs, which displayed a toroid-to-crumpled shape transformation for sufficiently large SPs. However, morphological differences between SPs containing ANFs or its monomer protein form (beta-lactoglobulin) were minor. Protein secondary structure of ANFs was somewhat affected by the spray drying process based on infrared spectroscopy. Transmission electron microscopy confirmed the homogeneous structure of SiNP-ANF SPs, while elemental mapping indicated even distribution of SiNPs and ANFs in the dry powder. The goal of understanding the effect of building block size, chemistry/softness (carried out in previous fiscal year), and aspect ratio on the morphology of ANF-infused spray-dried SPs was therefore completed.

ANFs also promoted the crumpling of SiNP-SPs when drugs were included in the spray-drying formulation. Drug dissolution experiments showed little variation in release kinetics when ANFs or beta-lactoglobulin was included in the SPs. Inhalation performance (cascade impactor) and nanoparticle release studies were not carried out due to large material requirements of the experiments owing to long ANF synthesis duration and high cost.
