Outline of Final Research Achievements

In the process of fibrillization, amyloidogenic proteins or peptides form droplets as an aggregation intermediate. The aim of this study was the structural study of the droplets. We employed chemical modification and took a stabilization strategy. We found intermolecularly hydrophobic interaction pivotal for controlling self-assembly and succeeded in gaining structural insight.

Academic Significance and Societal Importance of the Research Achievements

アミロイドーシスは、本来溶けて機能を発揮するタンパク質が不溶化しアミロイドとなり沈着することで発症される。この繊維化過程において液液相分離を経ることが近年明らかになりつつある。したがって、中間体である液滴(ドロプレット)に関する構造的な理解は、その後の疾患を予防できることや、現象の解明に大きく貢献できると期待される。
