Outline of Final Research Achievements |
Laser irradiation of PDL-MSCs at 6 J/cm2 and 8 J/cm2 significantly promoted proliferative activity without cell injury, and ALP activity was significantly increased 3 days after irradiation at 6 J/cm2 compared with the non-irradiated group. The calcified area of PDL-MSCs cultured in osteoblast differentiation induction medium was significantly increased after 3 weeks of weekly irradiation at 6 J/cm2 and 8 J/cm2, respectively. In addition, laser irradiation increased the expression of osteoblast differentiation marker genes in PDL-MSCs. Comprehensive analysis by RNA-seq 6 hours after irradiation revealed 45 differential expression genes (DEGs) at 3 J/cm2, 111 DEGs at 6 J/cm2, and 155 DEGs at 8 J/cm2.

Academic Significance and Societal Importance of the Research Achievements |
現在一般的に臨床応用されている、サイトカインや成長因子などを応用した歯周組織再生療法は、再生の範囲に限界がある。歯周組織再生療法の鍵として注目されるヒト由来歯根膜組織由来間葉系幹細胞(PDL-MSC)は、ヒト他家移植による安全性と有効性も確認され、既に他家移植のための細胞バンクも樹立されているが、細胞の状態により再生に差異があり、より再生に適したPDL-MSCの作出が求められている。 本研究によって、レーザーの光エネルギーによるphotobiomodulation効果がPDL-MSCの再生能へ与える影響の一端が明らかになり、光エネルギーを応用した新たな歯周組織再生療法開発の糸口とできる。
