Research Abstract

DNAおよびRNAの前駆体であるヌクレオチドは放射線や生体内外の環境ストレスにより多様な化学修飾を受けるが、その放射線生物影響における意義は不明で、早急に解決すべき課題ある。本研究では、環境ストレスによる修飾ヌクレオチドの生成が引き起こす生体障害を「ヌクレオチドプールの恒常性の破綻」としてとらえ、以下のアプローチで多様な放射線生物応答の制御機構を明らかにする目的で研究を進めている。
[1]放射線照射により生成される修飾ヌクレオチドをHPLCおよびLC-MS/MS法で分離、同定する方法を確立した。非放射性同位元素で標識した修飾ヌクレオチドを調製し、これを内部標準とするLC-MS/MS法による定量法を確立した。これらの方法を用いて、in vitroでの修飾ヌクレオチド生成モニタリングとin vivoの修飾ヌクレオチド検出系の確立を進めている。
[2]細胞外に放出・分泌される修飾ヌクレオチドとして、ATPの酸化体(2-OH-ATP)に注目し、細胞への影響の解析を進め、Bystander effectsのメディエータとしての可能性を追求している。
[3]修飾ヌクレオチド分解酵素を網羅的に探索する目的で、新規修飾ヌクレオチドを調製した。
[4]DNA中の修飾ヌクレオチド修復酵素を網羅的に探索する目的で、アッセイ系の確立を進めた。
[5]2つの新規ヌクレオチドプール浄化酵素欠損マウス胚性幹細胞を樹立した。
[6]神経変性の病態モデルマウスとして3xTG-ADマウスとSOD-TGマウスを導入し、DNA修復酵素及びヌクレオチドプール浄化酵素欠損マウスとの交配を進めている。
