| Research Abstract |
内泌撹乱化学物質・ビスフェノールA(BPA)の低用量作用が懸念されるなか、胎児(仔)期の脳神経成長に影響する可能性が判明した。本研究では、まず、この分子機構を解明することを目的とする。一方、BPAの代替としてビスフェノールAFなどを原料とする新プラスチックが続々と開発され、生産量が急増している。こうしたなか、これらの内分泌撹乱作用が心配されている。我々は、例えば、ビスフェノールAFがエストロゲン受容体α型にアゴニスト、β型にアンタゴニストとして強力に働くことを発見した。そこで、本研究ではこうした新世代ビスフェノールの核内受容体を介したシグナル毒性、内分泌撹乱作用の分子機構の解明にも取組む。
本年度、BPA受容体ERRγについてマウス胎仔脳でのChIP-Seq解析を実施した。その結果、ERRγを含め、48種のヒト核内受容体のうち、実に27種がERRγの転写制御を受けていることが判明した。そのうち、神経成長に関わる核内受容体2種が同定された。BPAの暴露の有無における発現リズムをマウス胎仔脳(日齢12～18)でリアルタイムPCR解析したところ、日齢が進むにつれ、暴露による変動が明らかとなった。一方、ショウジョウバエの培養細胞BG2-c6についてHiCEP解析を実施したところ、BPAの投与でmRNAの発現が減衰する核内受容体3種が確認された。＼
BPAが結合するERRγ以外の核内受容体を探索した。約20種で検討した結果、唯一、新たにCARが高親和性受容体として見出された。一方、BPAがショウジョウバエの多動性障害を誘因し、時計タンパク質PERの特定構造が関与していることが初めて分かった。ところで、ビスフェノールAFのERαおよびERβに対する結合様式がコンピュータモデリングで明らかになったが、向きが異なる単純な差違であり、これがどうしてアンタゴニスト活性を起こすのか?が新たな研究課題となった。
