Research Abstract

(1) TWEAKの受容体であるFn14が種々のヒトがん細胞に高発現していることを見出した。さらに、ヒト及びマウスFn14に対する抗体(ITEM4)にゲロニン毒素を結合し、ヒトメラノーマを移植したヌードマウスにおいて毒性無く強い抗腫瘍効果を認めた。(2) B16メラノーマやCT26大腸がんが産生するIL-18はNK細胞のPD-1発現を誘導して転移促進に働き、PD-1に対する阻害抗体によって転移が抑制されることを見出した。(3) MC38大腸がんや3LL肺がんにおいて、腫瘍内マクロファージが産生するMFG-E8が、がん幹細胞の維持と抗がん剤耐性及び樹状細胞の不活化に関わることを見出し、MFG-E8あるいはその受容体であるαVインテグリンに対する阻害抗体が単独あるいは種々の抗がん剤との併用で抗腫瘍効果を発揮することを示した。(4) AT-3乳がんを標的とした放射線療法との組み合わせにおいて、4-1BBに対するアゴニスト抗体とPD-1に対する阻害抗体の併用が最も効果的にCD8 T細胞依存性の抗腫瘍効果を増強することを示した。(5) 腫瘍内に浸潤している樹状細胞やマクロファージに高発現しているTIM-3は、アラーミンであるHMGB1を介した細胞外核酸の取り込みによるこれらの細胞の活性化を抑制しており、TIM-3に対する阻害抗体の投与によって、CpG-ODNやpoly(IC)といった核酸アジュバントの抗腫瘍効果やDNAワクチンの効果、さらには、化学療法によって生じる死細胞の免疫原性をも高めうることを見出した。(6) BRAF-V600E変異によるマウスメラノーマモデルにおいて、BRAF阻害剤PLX4720の抗腫瘍効果はCD8 T細胞依存性であり、4-1BBに対するアゴニスト抗体で相乗的に増強されることを示した。
