藻類における雌雄非対称性の成立とオルガネラ遺伝の方向を決める形態と構造の探索

Research Abstract

免疫サブトラクション法の再現性を検討するとともに、正逆のサブトラクションで雄(♂)の接合装置に特異的な抗体の調製に挑戦し、それをプロテオーム解析するための基盤を構築した。
1.ヒラアオノリの培養方法を再検討して雌雄配偶子の大量供給を可能にした。このことで雌雄配偶子のESTライブラリー構築、ゲノムシーケンス、プロテオーム解析が可能となった。
2.ヒラアオノリのマイクロサテライトマーカー、葉緑体とミトコンドリアのDNAマーカーを取得することができ、葉緑体とミトコンドリアの遺伝様式の大規模解析を続行している。
3.免疫サブトラクション法で配偶子の細胞表層に接合装置の性的二形性を検出している。アオノリに限らずノリの葉状体タンパク質の電気泳動は極めて難しい。これを克服する方法を考案し、ウェスタンブロット解析などが容易になった。免疫サブトラクション法を拡張して、抗雌(♀)抗血清と抗雄(♂)抗血清を調整して、雌(♀)では200kDa、雄(♂)では40kDaの特異抗原を見いだすことができた。また、雄(♂)に特異的な60kDaと40kDaのバンドも検出した。北大グループと共同で、野外採集した褐藻の配偶子でも調査予定である。
4.抗雌(♀)抗血清と抗雄(♂)抗血清を用いて蛍光抗体染色をおこない、雌雄に特異的な細胞構造の発見に務めている。接合装置だけでなく雌雄の膜が融合する領域の染色にも成功した。FE-SEM免疫電顕に関しては筑波大グループと共同研究する予定である。
5.雌雄配偶子で67～69万リード規模のESTを構築した。また、雌雄配偶子で77～95万リード数程度ではあるが全ゲノム解析を行った。さらに、共同研究で、雌雄配偶子の細胞膜分画をマスでプロテオーム解析した。いずれも限られた規模なので、3つのデータをそれぞれ補完することで、雌雄特異的な遺伝子群の同定を可能にした。

Reason

本研究課題の「オルガネラ遺伝の方向性」と「雌雄配偶子の非対称性」を調べるためには、1)マイクロサテライトマーカー、葉緑体とミトコンドリアのDNAマーカーを取得と2)免疫サブトラクション法の再現性調査が欠かせない。この両方ともにクリアできたので、達成度はおおむね順調に進展していると考えられる。

Strategy for Future Research Activity

免疫サブトラクション法の再現性をヒラアオノリの異なる株と褐藻などの他の藻類で見る必要がある。調製された雌雄特異的な抗血清の利用方法についても検討する必要があろう。プロテオーム的な解析とリンクさせるためには、雌雄特異的な抗血清を用いた免疫沈降法によるマス解析が是非とも必要になろう。
