両生類と爬虫類に生じたゲノム・染色体再配列過程の分子細胞遺伝学的検証

Research Abstract

脊椎動物では、その進化過程において2回のゲノム重複が生じたと考えられている。しかし、ゲノム倍数化後のゲノム・染色体再編成の過程はほとんどわかっていない。我々は、約4千万年前にゲノム倍数化が生じた異質四倍体種であるアフリカツメガエル(Xenopus laevis) (2n=36)を対象として、cDNAクローンを用いたFISH法によって60の遺伝子からなる染色体地図を作製し、二倍体種のネッタイツメガエル(X. tropicalis) (2n=20)の染色体地図と比較した。その結果、9組の同祖染色体を同定し、60遺伝子中50遺伝子(83%)において四倍性が保存されていることを明らかにした。また、同祖染色体間での相同性は極めて高く、染色体間での転座はなく、2組の同祖染色体で逆位が検出されただけであった。これらの結果から、倍数化後のゲノムの二倍体化の過程において、染色体の再編成は必須ではないことを明らかにした。
鳥類の核型は、サイズが大きく異なるマクロ染色体とマイクロ染色体で構成され、両者のGC含量は大きく異なることから、染色体サイズ依存的なゲノム構造の区画化が存在する。爬虫類は同様の核型をもち、鳥類との染色体の相同性は極めて高いが、ゲノム構造の区画化の有無については良くわかっていない。我々は、シマヘビ(Elaphe quadrivirgata)を用いて、マクロ染色体とマイクロ染色体にマップされた機能遺伝子の各コドンの3番目の塩基のGC含量(GC3)を比較した。その結果、マイクロ染色体の遺伝子は有意にGC3含量が高く、一方、核型進化の過程でマクロ染色体に挿入された、祖先核型のマイクロ染色体由来の遺伝子では、GC3含量が高くなる傾向が見られた。これら結果は、爬虫類と鳥類の共通祖先である竜弓類において、すでに染色体サイズ依存的なゲノム構造の区画化が存在していたことを示している。
