Research Abstract

(1)CD22(Siglec-2)高親和性リガンドの開発とB細胞免疫応答の制御:
B細胞表面に特異的に発現するシアル酸認識レクチン(CD22)の高親和性リガンド(GSC-718)のin vivo免疫賦活試験(連携研究者・鍔田武志らとの共同研究)を遂行するため、大量合成法の確立とアッセイサンプルの生体適合性について検討した。その結果、化学合成品におけるリチウムイオンの混在が活性を低下させることが明らかとなった。一方、GSC-718をリードとした構造活性相関研究を行うため、一連のシアル酸誘導体の合成に成功した(Synthesis, 18, 2968-2974, 2011 ; Current Medicinal Chemistry, 18, 3537-3550, 2011)。
(2)ガングリオシド合成法の革新と応用:
グルコシルセラミド誘導体を最終のグルコシル化アクセプターとするカセット合成法と、新しいシアル酸連結法により、一連の棘皮動物由来新奇ガングリオシド類の世界初全合成を達成した(Chem.Comm., 47, 9726-9728, 2011 ; Chem.Eur.J., 17, 5461-5651, 2011 ; Angew.Chem.Int.Ed.Engl.50, 2330-2333, 2011 ; Chem.Eur.J.17, 588-597, 2011)。
(3)1分子イメージング用蛍光ガングリオシドの開発と応用:
ガングリオシドGM3とGM1の蛍光標識化に成功し、細胞膜ラフトのマーカー分子としての物理化学的評価を行った。その結果、糖鎖非還元末端への蛍光標識化が1分子イメージング解析に有効であることが明らかとなり、実際、P24細胞膜上においてラフトマーカーとして機能することを確認した。

Reason

CD22(Siglec-2)高親和性リガンドの開発は順調に進み、国際誌Synthesis, Bioorg.Med.Chem.,及びCurrent Medicinal Chemistryに論文掲載済み。さらに、棘皮動物由来新奇ガングリオシド類の世界初全合成の達成は、高インパクトファクター誌Chem.Comm., Chem.Eur.J., Angew.Chem.Int.Ed.Engl.に連続して掲載され、計画以上の成果を得た。

本研究で目標としている6つの課題は、いずれも順調に進展しており、計画通り遂行する予定である。
