Research Abstract

(1) 新戦略に基づくガングリオシド合成法の革新と応用：グルコシルセラミド誘導体を最終のグルコシル化アクセプターとするカセット合成法と、新しい修飾シアル酸連結法を用いて、アオヒドデ由来ガングリオシドLLG-3及びムラサキヒトデ由来ガングリオシドGAA-7の世界初全合成を達成した(Organic Lett., 14, 6342-6345, 2012)。これらは強力な神経突起伸展活性を有しており、医学的応用が期待される。
(2) 自己免疫性神経疾患に関係する新奇ガングリオシドの全合成：筋萎縮性側索硬化症（ALS）に類似する自己免疫性神経疾患の原因物質として単離されたガングリオシドX2の世界初全合成を達成した（Chem. Asian J., 7, 1041-1051, 2012）。これらは、GBSの診断に重要である。
(3) シアロ糖鎖を足場とするウィルス感染機構の解明:ヒトメルケル細胞癌を惹起するポリオーマウィルス（MCPyV）の感染機構を解明するために、MCPyVのキャプシド蛋白VP1と合成シアロ糖鎖の結合特異性を糖鎖マイクロアレイ及びX線により解析した。その結果、VP1はNeu5Ac-α2,3-Gal構造を認識して結合・感染することが明らかとなった(PLoS Pathogens, 8, 1-11, 2012)。
またインフルエンザウィルスの新規ガングリオシド受容体の全合成に成功した（Molecules, 17, 9590-9620, 2012）。

交付申請書に掲げた6つの研究目的について大いに研究を推進し、この3年間で計22報に上る国際的英文誌にその成果を報告・公開した。全て世界で初めて達成されたレベルの高い研究成果であり、国内外において高く評価されている。
