PSGL-1受容体を介するエンテロウイルス71感染・病原性発現の分子基盤

Research Abstract

我々は、手足口病および重症中枢神経合併症の原因ウイルスであるエンテロウイルス71(EV71)の白血球細胞における機能的受容体分子としてP-selectin glycoprotein ligand-1(PSGL-1)を同定した。EV71感染初期過程および病原性発現の分子基盤の解明のため、EV71感染増殖および病原性発現におけるPSGL-1受容体の機能について解析し、以下の結果を得た。
1. PSGL-1結合性・非結合性EV71株のカプシド領域アミノ酸の比較解析に基づく感染性クローンへの特異的変異の導入により、PSGL-1結合性を規定する主要なアミノ酸部位としてVP1-145を同定した。VP1-145アミノ酸置換によりEV71のPSGL-1結合能とPSGL-1依存的ウイルス増殖能が規定されることを明らかにした。クライオ電顕等を用いたEV71粒子の構造解析により、EV71中和活性を有するモノクローナル抗体の結合部位が、PSGL-1結合領域と一部重複することを示した。
2. PSGL-1受容体依存性および非依存性EV71感染・病原性発現機構を解析するため、PSGL-1結合に関与するアミノ酸のみが異なる感染性クローン由来PSGL-1結合株(02363-EG株)とPSGL-1非結合株(02363-KE株)を調整し、ウイルス学的性状を解析した。PSGL-1結合株および非結合株を、それぞれ、カニクイザルに感染し、病原性発現、異なる組織におけるウイルス増殖、ウイルス抗原・病変の局在、免疫炎症反応、個体内でのウイルス遺伝子変異等について比較検討した。PSGL-1非結合株はPSGL-1結合株と比較して、感染個体内でよく増殖し、無菌性髄膜炎等の臨床症状も強い傾向が認められた。PSGL-1結合株は感染後すみやかにPSGL-1結合領域アミノ酸に変異を生じることが明らかとなった。
