Research Abstract

【目的】TDP-43はN末端側RNA結合ドメインとC末端側タンパク結合ドメインを有する核タンパクある。ALS/FTD脳では、TDP-43が異常リン酸化・ユビキチン化・C末端切断の修飾を受け細胞質へ移行し、封入体を形成する。しかしTDP-43の生理的な機能は明らかでない。最近Freibaumらは質量分析によりヒトTDP-43結合タンパク数百種類を同定した。SephtonらはRIP-seq解析によりラットTDP-43結合RNA数千種類を同定した。本研究ではこれらの網羅的解析データを用いて、TDP-43標的遺伝子と結合タンパクの分子ネットワークの生物学的意味付けを行った。【結果】ラットTDP-43標的遺伝子(n = 4163)の分子ネットワークは、axon guidance, RNA post-transcriptional modification, transcriptional regulation by p53と、ヒトTDP-43結合タンパク(n = 227)の分子ネットワークは、ribosome, spliceosome, protein synthesis, spliceosome assemblyと密接に関連していた。ラット遺伝子IDをヒトに変換しヒトTDP-43標的遺伝子(n = 4063)と結合タンパク(n = 227)を統合した包括的分子ネットワークを解析すると、ribosome, axon guidance, spliceosome, RNA-binding, RNA post-transcriptional modification, transcriptional regulation by p53と関連性を認めた。【結論】TDP-43は核ではspliceosomeと細胞質ではribosomeと密接にリンクし、神経細胞機能維持を制御していると考えられた。
