ナノ粒子による脳疾患治療への挑戦

Research Abstract

平成23年度は初年度の評価を基に次のように検討を行った。
i)投与経路の異なる粒子薬物動態解析:静脈、動脈、経口、経鼻、経肺や頭頂後頭溝への直接投与など、さまざまな投与法によるナノ粒子の取り込み評価を行った。ICRマウスを中心に検討を進め、ラジオラベリング法(125I,RI)とともに電子スピン共鳴法(ESR)により経口投与では少なくとも1%程度の取り込みが確認された。特に経口投与によっても脳への取り込みが確認され、今後の脳疾患に対するナノ治療に期待される。
ii)脳梗塞再灌流障害に対するナノ粒子取り込みに関する研究:脳梗塞再灌流障害は血液脳関門の損傷をきたすと報告されているものの、その真偽に関しては定かではないため、本研究ではカテーテルによる脳動脈閉そくに伴う虚血後、カテーテルを除去することにより再灌流を行った後、ナノ粒子を投与し、その取り込みに関して検討を行った。特に梗塞障害部位以外の取り込みを確認するため、蛍光ラベルナノ粒子を作成し、検討を行い、梗塞から離れているところでの粒子の存在を確認した。またESRによる取り込みも確認した。取り込み量は少なくとも1%以上に達し、効果的な取り込みが確認された。
iii)収束超音波ビームによる血液脳関門透過に関する解析:集束超音波ビームによる血液脳関門透過研究を行い、透過量が10倍程度にあがることを確認した。収束超音波ビームはその強度により出血画餅発するため、その調整が重要であることを確認した。
