機械的刺激による血管弾性線維形成機序の解明

Research Abstract

血管弾性線維は動脈の構造を保つ上で極めて重要な要素であり、その破綻は老化や血管病と密接に関係している。弾性線維は一般に細動脈から大動脈へと圧負荷が増加するにつれて発達する。従って、血管特有のなんらかの機械的刺激(血流や圧など)が弾性線維形成に影響を及ぼすことが推定されるが、これまでに弾性線維形成を定量的に評価するための良い実験系がなかったことなどから、いまだに不明な点が多い。そこで本研究では機械的刺激(血流や圧など)の種類や強度などの変化が血管弾性線維形成に及ぼす影響について解明することを目的に研究を行った。本年度は平滑筋培養細胞の積層化技術を用いて、ラット胎仔から採取した大動脈血管平滑筋細胞を順次積層化し、7層まで細胞生存性を保ったまま、重層化することに成功した。この大動脈血管平滑筋細胞層の再構築系を用いて、血管弾性線維形成に影響を与える要素の検討を行った。その結果、1)血管平滑筋培養細胞は単層に比べて重層化する方が血管弾性線維の形成が促進されること、2)細胞培養液のウシ胎仔血清濃度を10%から1%へ低下させる、もしくは血清を除去すると血管弾性線維の形成が促進されること、3)プロスタグランジンE刺激を行うと血管弾性線維の形成が有意に抑制されること、4)血管平滑筋培養細胞への伸展刺激を繰り返すと血管弾性線維の形成が促進されること、を見出した。本結果に基づき、より定量的な解析のための準備を整えることが出来た。
