| Research Abstract |
かゆみ惹起神経ペプチドであるサブスタンスP(SP)刺激によってコレシストキニン受容体であるCCK2発現元進がみられたことは、CCKがCCK2Rシグナリングを介して、負の制御としてかゆみ抑制作用を示す可能性を示唆した。ICRマウスの背部を剃毛し、そこにSP(100nm/部分)を皮下注し、マウスの後肢掻破行動を誘導した。SP皮下投与10分前に剃毛部に種々のCCKペプチドを塗布し、溶液のみのコントロールと比べたところ、掻破回数がとくにCCK8Sで減弱した。CCK皮下投与でも同様の効果をもたらすか、SPと同時にCCK8Sを皮下投与したところ掻破回数が減弱した。皮下投与でもかゆみ抑制効果を示すのであれば、角化細胞以外に肥満細胞も標的細胞となることが示唆された。マウス胎仔由来肥満細胞をcompound 48/80で脱顆粒刺激する系で、CCK8Sを添加したところ脱顆粒は抑制された。以上のことから、CCKはかゆみを抑制する能力をもち、少なくとも肥満細胞を機能を抑制して効果を示すのではないかと考えられ、創薬への端緒となった。
