細胞挙動コントロールを可能にする導電性ソフトインターフェイスの構築

Research Abstract

新規チオフェンモノマーを調製し、トップダウン/ボトムアップ双方のアプローチを展開した。
ポリチオフェンによる層構造薄膜 交互積層法を応用した電解重合により、層構造を有する導電性ポリチオフェン薄膜を調製し、表面特性のオルタナティブな変化を可能にした。モジュレーション、バイオコンジュゲーションに機能するそれぞれのモノマーの組成を検討、最適化した。電解重合溶液中の双性イオン性モノマーの存在比により、薄膜の親疎水性を制御し、細胞の接着及び、生体分子の非特異吸着をコントロールした。導電性高分子を用いることで、交互積層法を応用した電解重合により機能性薄膜を調製し、その特性の迅速な転換を可能にした。細胞接着性に、機能性薄膜表面の親疎水性が与える影響を明確にした。
トップダウン/ボトムアップの組合せによる細胞パターニング フォトリソグラフィ(トップダウン)と、交互積層法を応用した電解重合(ボトムアップ)を組合せ、パターン化した表面上での細胞行動コントロールを成し遂げた。細胞接着に有効なドメインと、非特異吸着に有効なドメインを複合し細胞のパターニングを可能にした。パターン化したデバイスと、目的の場所にのみ接着した細胞は、7日間に及ぶ培養期間を経た後にも安定であった。
導電性高分子ナノ構造による細胞捕捉の高効率化 導電性高分子によるナノ構造プラットフォームをターゲット細胞捕捉の高効率化に応用した。ナノドットを電解重合により、サイズ、密度を制御し調製した。ナノドットのカルボキシル基を用いて、循環腫瘍細胞の抗原に対する抗体で修飾し、ターゲット細胞捕捉の高効率化を達成した。平滑な薄膜表面と比較し、ナノドット表面では約5倍の特異的な捕捉が可能であった。この方法の有意な点は、簡便なナノ構造の調製法と、官能基の有用性、力学物性であり、これらの特長はがん診断、循環腫瘍細胞の検出分野での応用に適している。

Reason

これまでに、交互積層法を応用した電解重合に成功し、電解重合による、材料特性の速やかな変更を可能にした。また、この方法とトップダウンアプローチであるリソグラフィーを組合せ、細胞の空間的なアライメントと挙動のコントロールを達成した。当初の目的に即し順調に進行しており、この研究に関する5報の査読付き論文を発表した。

Strategy for Future Research Activity

導電性高分子の利点は、電気刺激もしくは電気シグナルなどの電気的なインフォメーションとの共存が可能な点である。この点を有効に利用し、多種の外的刺激に対し応答性を有する、ポリチオフェンによるスマートなマテリアルを創出し、細胞との相互作用に応用する。これら導電性ソフトマテリアルが、多様な刺激の複合的な影響を精査する有効なプラットフォームとなることが期待される。このプラットフォームの活用により、細胞工学における外的刺激が及ぼす影響の系統的な理解が可能となる。
